IMP3 RNP safe houses prevent miRNA-directed HMGA2 mRNA decay in cancer and development.
The IMP3 RNA-binding protein is associated with metastasis and poor outcome in human cancer. Using solid cancer transcriptome data, we found that IMP3 correlates with HMGA2 mRNA expression. Cytoplasmic IMP3 granules contain HMGA2, and IMP3 dose-dependently increases HMGA2 mRNA. HMGA2 is regulated by let-7, and let-7 antagomiRs make HMGA2 refractory to IMP3. Removal of let-7 target sites elimina...
متن کاملModifiers of solid RNP granules control normal RNP dynamics and mRNA activity in early development
Ribonucleoproteins (RNPs) often coassemble into supramolecular bodies with regulated dynamics. The factors controlling RNP bodies and connections to RNA regulation are unclear. During Caenorhabditis elegans oogenesis, cytoplasmic RNPs can transition among diffuse, liquid, and solid states linked to mRNA regulation. Loss of CGH-1/Ddx6 RNA helicase generates solid granules that are sensitive to m...
متن کاملBioinformatics identification of miRNA-mRNA regulatory network contributing to lung cancer invasion
Background: Over the past 15 years, significant insights have been gained into the roles of miRNAs in cancer. In various cancers, miRNAs can act as oncogenes, tumor suppressors, or control the metastasis process by modulating the expression of numerous target genes. This study is aimed at determining molecular network of miRNA-mRNA regulating lung cancer invasion, by bioinformatics approaches. ...
متن کاملBioinformatics Identification of miRNA-mRNA Regulatory Network Contributing Primary Lung Cancer
Introduction: In clinical practice, distinguishing invasive lung tumors from primary tumors remains a challenge. With recent advances in understanding biological alterations of tumorigenesis and molecular analytic technologies, using these molecular alterations can be sensitive and tumor-specific as biomarker for the stratification of patients. In this study, the molecular network of miRNA-mRNA...
متن کاملIntegrative Analysis of miRNA-mRNA and miRNA-miRNA Interactions
MicroRNAs (miRNAs) are small, noncoding regulatory molecules. They are involved in many essential biological processes and act by suppressing gene expression. The present work reports an integrative analysis of miRNA-mRNA and miRNA-miRNA interactions and their regulatory patterns using high-throughput miRNA and mRNA datasets. Aberrantly expressed miRNA and mRNA profiles were obtained based on f...
